제목 One step purification and immobilization of enzymes using Poly(2-acetamido acrylic acid) PAAA hydrogel
초록 We developed one-step purification and immobilization system of enzymes using metal-complexed poly(2-acetamido acrylic acid) (PAAA) hydrogel. Directly from cell lysate, polyhis-tagged recombinant enzymes (Glutamyl aminopeptidase) were purified and immobilized to the PAAA hydrogel via the chelated metal ion affinity interaction. PAAA hydrogel was synthesized using 2-Acetamidoacrylic acid(AAA) and 2,2´-[(1,4-dioxo-1, 4-butanediyl)diimino] bis(2-propenoic acid) in DMSO by potassium persulfate(KPS). For the preparation of the Ni2+ -complexed PAAA hydrogel(Ni2+ -PAAA hydrogel), the PAAA hydrogel was immersed in solution of 0.1M nickel chloride(NiCl2) and then maintained under stirring at room temperature for 24h. In the presentation, the enzyme binding capacity, enzymatic activity, and reuse and storage stability of immobilized enzyme will be discussed.
