제목 | Preparation and biological effect of gelatin-based scaffolds for cultured artificial dermal substitute |
초록 | Skin substitutes with living cells have been promptly produced by in vitro three-dimensional culture techniques. These techniques promise a new approach to the repair and reconstruction of tissue damaged by burn injury, mechanical injury and pressure sores. The denatured type of collagen, gelatin has been used widely in medicine as wound dressing, adhesive and absorbant pad for surgical use because collagen and degradation products of collagen may be attractants for fibroblasts in vivo during wound repair, fracture healing and embryogenesis. The objective of the present study was to prepare the porous gelatin-based scaffolds using two kinds of preparation method such as freeze-drying and salt-leaching and to investigate their cell attachment, morphological characteristics and wound healing effects. In experiments, scaffolds were manufactured by freeze-drying and salt-leaching method, respectively, and crosslinked by 1-ethyl-(3-3-dimethylaminopropyl) carbodiimide hydrochloride (EDC). The scaffolds showed the inter-connected pore structure of which average pore size was about 100~300μm. In biodegradation in vitro, the degradation rate increased with porosity of scaffold. Also, it was found that gelatin-based sponge has enough mechanical properties for cell culture and transplantation for artificial skin. Results on cell attachment revealed that the high gelatin content was suitable for cellular attachment and distribution in three-dimensional fibroblast culture. |
