제목 Vacuum-dry method for the improvement of release kinetics of BCNU from BCNU-loaded PLGA wafer
초록 1,3-bis(2-chloroethyl)-1-nitrosourea (BCNU) has been used as an antineoplastic drug for the treatment of brain tumor. However, BCNU has short half-life and side effects in vivo. Poly(D,L-lactide-co-glycolide) (PLGA) wafers offer many advantages such as controlled drug release rate, good biocompatibility and biodegradable property. However, mechanical mixing is not suitable for sensitive BCNU, which can be changed its chemical property by exposing on air and/or temperature and so on. Therefore the aim of this work is to obtain uniform compound without an influence of external factors as mentioned above. Dissolved PLGA in solvent could be recrystallized by vacuum dry and recovered as powder form with each different physical property. BCNU was mixed with the obtained PLGA and the wafer was fabricated by compression molding method and the size of the wafer was 3 × 1 mm. In vitro release of BCNU from the wafer was assayed by high performance liquid chromatography (HPLC). Morphological observation was performed by scanning electron microscopy (SEM). Degradation studies such as mass loss and water uptake were tested. The pre-mixed BCNU loaded PLGA particles were characterized by X-ray diffractometer (XRD) and differential scanning calorimetry (DSC), respectively. As a result of vacuum-dry method, the PLGA powders became new shape. And it was derived to nearly zero-order release profiles
