제목 Effect of Crosslinked Alginate Films on Proliferation and Phenotype of Corneal Endotherial Cells
초록 To evaluate the percentage of endothelial cell damage induced during a surgical technique of Descemet's membrane separation containing healthy endothelium. Alginate is a polysaccharide block copolymer consisting of repeats of -D-mannuronic acid and R-Lguluronic acid monomer. A new method is identified here for producing Ca2+ cross-linked thin films of sodium alginate, using an 0.4, 0.6, and 0.8M CaCl2 solution. Corneal endotherial cells(CEC) were seeded into 1% alginate films. After 1, 3 and 7days of cell seeding, cell proliferation and viability were measured via MTT assay. RT-PCR was conducted to confirm mRNA expression of ColⅤfor CEC marker. We confirmed that cell viability on alginate films of 0.6M were higher than other films. Also, RT-PCR and histology staining results, alginate films of 0.6M showed that the improved gene characteristic of CEC cells.
