Synthesis and Self-assembling Properties of Poly(ethylene glycol)/poly(ε-caprolactone) AB2 Miktoarm Copolymers for Anticancer Drug Delivery |

Amphiphilic AB2 miktoarm copolymers that mimic structural characteristics of phospholipid show a high potential for forming nanostructures and application as an efficient drug carrier system. In this study, a series of new amphiphilic AB2 miktoarm copolymers consisting of hydrophilic PEG block and hydrophobic PCL block were synthesized. The chemical compositions of PEG-PCL2 miktoarm copolymers were confirmed by 1H-NMR, GPC, and FT-IR measurements. The PEG-PCL2 miktoarm copolymers with relatively short PCL blocks readily formed spherical micelles when they self-assembled in aqueous solution. However, the worm-like micellar structure was observed when the length of PCL blocks increased. Dxorubicin (DOX) was efficiently loaded into PEG-PCL2 miktoarm nanocarriers. In vitro release profiles of Dox-loaded self-assembled nanocarriers showed sustained release patterns. PEG-PCL2 miktoarm nanocarriers exhibited enhanced cellular uptake into cells compared to Dox. |
