We demonstrate the nanostructured microparticles (NM) containing a mucoadhesive polymer for enhanced preocular retention to consider them as potential drug carrier to the eye in this work. The NM are each composed of the entangled nanofibers to give an enlarged specific surface area and thus, can better adhere to the preocular mucus surface. With this physical means of microparticle design, the microparticles can still be composed mainly of a wall-material, poly(lactic-co-glycolic acid), eventually needed for controlled drug delivery, while the effect of an additive, mucoadhesive material, polyethylene glycol, can be synergistically improved with the nanostructured morphology. Thus, when formulated in a dry tablet dosage form, the NM in this work show more than a 10-fold increase in preocular retention in vivo, as compared with the conventional spherical microparticles. |
