제목 | Effect of glycolide and lactide monomers on the release profiles of BCNU from BCNU-loaded PLGA wafer |
초록 | 1,3-bis(2-chloroethyl)-1-nitrosourea (BCNU) has been used as a chemotherapeutic agent for brain tumor. Small sized BCNU-loaded PLGA wafer has been developed to insert into tumor bearing rat model for in vivo test. Accordig to a wafer size, release rate was affected and the small wafer must represent the same release profiles as the wafer which is used for clinical trial. Poly(lactic-co-glycolic acid) (PLGA) is biodegradable polymer composed of lactide and glycolide monomers which are also biodegradable monomers. In our present study, the effect of glycolide and lactide monomers on the release profiles of BCNU from BCNU-loaded PLGA wafer, repectively was assayed by high performance liquid chromatography (HPLC) at 248㎚. Also, change of media pH was measured and surface and cross section of wafer morphology was observed by scanning electron microscopy (SEM) during release period. Release rate was increased with increasing amount of glycolide and lactide monomers, respectively due to increasing porosity and channels in matrix. So the release profiles could be controlled and the formulation showing near zero-order release was obtained. Acknowledgment: This work was supported by KMOCIE |
