제목 Evaluation of in vitro release profiles of BCNU from multi-layered PLGA wafer containing glycolide monomer
초록 With the growing need for optimization of therapy, matrix systems providing zero-order release than typical first-order release are becoming more important. But poly(lactic-co-glycolic acid) (PLGA), one of the biodegradable polymers and used in this study, has diffusional controlled release, bulk erosion property, and non-linear release behaviour. In this study, we have established the multi-layered matrix system for controlled release and used glycolide monomer as additive to diminish long-term release. The multi-layered PLGA wafers consist of core and barried layer with 3.85% BCNU and different ratio of glycolide monomer. In vitro release profile of BCNU from multi-layered PLGA wafer was assayed by high performance liquid chromatography (HPLC) and morphological observation was performed by scanning electron microscopy (SEM). From results of this study, we could control the release profile of BCNU from multi-layered PLGA wafer depends on the ratio of glycolide monomer in core and barrier layer. In our experiment, multi-layed PLGA wafer consisted of 60% glycolide monomer in core layer and 40% glycolide monomer in barrier layer showed almost zero-order release profiles.
