Recently, photodynamic therapy (PDT) has been intensively studied and employed as a powerful non-invasive technique for various cancer treatments. Particularly, near infrared (NIR) photothermal therapy (PTT) using gold nanoparticles is a promising hyperthermia strategy. In this study, AuNR-photosensitizer conjugates with glutathione-sensitive linkages have been successfully developed. Photosensitizer is used for photodynamic therapy and AuNRs are used as a hyperthermia agent for photothermal therapy. A series of AuNRs with different aspect ratio were synthesized by seeded growth method. Then, ligand-conjugated block copolymers were employed to modify AuNRs. The physicochemical properties, singlet oxygen generation, and photothermal activities of the AuNRs-based system were evaluated. In vitro cellular uptake and phototoxicity test were also demonstrated using MCF 7 and A549 cell lines. |
