Thermosensitive META-Pluronic-Glycolide-Heparin Hydrogel for proliferation and differentiation of adipose-derived stem cells |

Pluronic is nontoxic poly(ethylene oxide)-(polypropylene oxide)-poly(ethylene oxide) (PEO-PPO-PEO) triblock copolymers that form a thermoreversible gel. It has found a wide variety of applications in tissue engineering and drug delivery. In this work, a functional Pluronic (F127) derivative, such as 4-methacryloxyethyl trimellitic anhydride(META), glycolide, and heparin-grafted Pluronic (F127-G5-META-Heparin) was synthesized and then the derivative was subjected to binding to transforming growth factor(TGF-β). The effects of the TGF-β-incorporated hydrogel tasted were in the chondrogenic differentiation, of adipose-derived stem cell (ASC). The cell-encapsulated hydrogels were cultured for specific times and evaluated for cell number, viability, cartilage-specific ECM synthesis, and gene expression levels of collagen type I and II. This Pluronic-based hydrogel system is expected to be useful as an injectable hydrogel for tissue regeneration. |
