제목 | Osteogenesis of Bone Marrow-Derived Mesenchymal Stem Cells in Porous Polymeric Scaffolds contained Natural Protein Powders |
초록 | Bone marrow contains a population of mesenchymal stem cells with the ability to differentiate into cells that from bone, cartilage, adipose, and other connective tissues. Stem cells can be isolated from bone marrow aspirates and expanded in vitro. Demineralized bone particles (DBP) contains bone morphogenietic protein (BMP), which is known to induce new bone formation at an extraskeletal site. Also small intestinal submucosa (SIS) contains many cytokines and specific proteins, which is known to induce bone formation and neurogenesis. In this study, we have characterized the osteogenic potential of MSCs in porous polymeric scaffolds, PLGA, DBP/PLGA and SIS/PLGA, to support osteogenic differentiation following dexamethasone and vitamin D3 treatment. We made porous polymeric scaffolds using DBP, SIS and PLGA by solvent casting / salt leaching method. In vitro, MSCs were seeded in polymeric scaffolds for 42 days. The osteogenesis of MSCs-seeded polymer scaffolds culture was assessed by histology and RT-PCR for osteogenic phenotype.
