제목 | Gentamicin Sulfate-loaded PLGA wafer for Antibiotic Local Delivery System |
초록 | Poly(D,L-lactid-co-glycolide) (PLGA) is biocompatible and biodegradable polymer used a wide range of controlled drug delivery application. Gentamicin sulfate (GS) is a higly water soluble antibiotic of the aminoglycoside group and widely used for the treatmet of osteomyelitis and the prevention of secondary infection after orthopedic surgery. GS was mixed with PLGA by freezer mill and the wafer was prepared by direct compress method. The objective of this study was to prepare the biodegradable wafer for sustained release of GS and PLGA mixture. The effects of wafer sizes and excipients were investigated. In vitro study was performed in pH 7.4 phosphate buffered saline at 37℃. Surface and cross sectional morphology were observed by scanning electron microscope (SEM). Drug content and release amount of GS were analyzed by high performance liquid chromatography (HPLC) and the released GS activity was observed by microbiological assay using Staphylococcus aureus.
