제목 | Effect of Semipermeable Membrane Materials on the Improved-Osmotic Tablet System for Nifedipine Delivery |
초록 | Osmotic pump tablet system for oral administration has many advantages such as constant delivery rate or minimized fluctuation of drug concentration in plasma and thereby reduced risk of adverse reactions, extended action period or reduced inconvenience of frequent re-dose and hence improved patient compliance, and in vivo predictability of release rate on the basis of in vitro data. For the improvement of release pattern and water-insolubility of nifedipine (ND), we prepared improved-osmotic tablet (IOT). ND is practically insoluble in water with solubility less than 10 g/ml, a well known and most widely used coronary vasodilat or from the group of dehydropyrine derivatives. The aims of study were to prepare IOT by novel mould device, and to investigate the effect of different preparation parameter, such as coating process. The coating solution was composed of cellulose acetate as membrane material, polyethylene glycol 200, triacetin, KCl, lactose and fructose as permeability controller, and acetone, ethanol and water as solvent. The release patterns and the membrane morphology of novel tablet were observed by high performance liquid chromatography and scanning electron microscope.
