Preparation and Characterization of Carboxymethyl-dextran based conjugates for Cancer Therapy |
초록 |
Clinically available taxanes such as paclitaxel and docetaxel are the most promising classes of anticancer agents for the treatment of various cancers. However, its therapeutic potential has been severely hampered by its poor solubility in water and indiscriminate cytotoxicity in normal cells. In an attempt to solve this problems, dextran derivatives have been extensively investigated for biomedical applications due to their excellent water solubility and biocompatibility. In particular, carboxymethyl dextran (CMD) have been prepared in which drugs can be released upon degradation of the polymers because of multiple functional group for chemical conjugations. In this study, we have prepared CMD-drug conjugates using facile chemistry, which could enhance the solubility of poor water-soluble anticancer drugs and selectively release the drugs in the acidic condition. The physicochemical characteristics of CMD conjugates were investigated using 1H-NMR, HPLC and Confocal microscopy. |
