제목 | Polymer Prodrug Approaches Applied to Paclitaxel |
초록 | Paclitaxel is a unique tubulin-interacting cytostaticum. Unlike other conventional antimitotic agents, which inhibit the microtuble disassembly process, paclitaxel is extremely insoluble in water and therefore difficult to apply. Paclitaxel has a special mechanism of action that inhibits SMC(smooth muscle cell) proliferation and migration. So, it has been applied to the surface treatment of stent for crown artery disease. But, restenosis and thrombosis after e.g.percutane coronar interventions are still problems in angioplasty. In this study, we intend to induce an excellent anti-thrombosis action and a new drug delivery system based on a harmonized hydrophobicity/hydrophilicity ratio by developing a drug-eluting stent having a water-soluble paclitaxel prodrug. It can be conducted to control a drug release rate competitively by incorporation of the appropriate length of PEG-chain and of the various self-immolating linkers onto the drug-eluting stent. We have synthesized and characterized water-soluble paclitaxel prodrugs with the appropriate length of PEG-chain and the various self-immolating linkers. |
