Immune Response of Purified and Non-purified Alginatein Three dimensional microcapsules |

Alginate, emains the most widely used biomaterial for immobilizing cells to be transplanted, because of the good viability of encapsulated cell and the relatively ease processing for cell encapsulation. However, the main drawback is the immune rejection in vivo. To overcome this problem, we have developed using modifed Korbutt’s method for alginate purification. T-PCR was performed to assess mRNA expression for inflammation cytokines such as IL-1, 1L-6. Purified and non-purified alginate microcapsules were implanted into a back of BALB/c-nu nude mouse, and the implanted alginate capsules were extracted after 1, and 2 weeks. Tissues surrounding implants were harvested and evaluated by histology through H&E staining and immunohistochemistry through ED-1 staining. This study demonstrates that purified alginate reduce inflammation reaction in vitro and in vivo. This research was supported by WCU (R31-20029) and MBC (0405-BO01-0204-0006). |
