제목 | Anti-bone Resorption of Pamidronate-loaded PLGA Wafer in Experimental Aural Cholesteatoma |
초록 | Pamidronate disodium (PD) is an important representative of the bisphosphonates, potently inhibit bone resortion and is used for the treatment of bone disease such as osteoporosis, hypercalcemia of malignancy and Paget's disease. In this study, PD is chosen as a model drug due to its lower autotoxicity than other bisphosphonates. PD was incorporated into poly(L-lactide-co -glycolide) (PLGA), drug carrier, for homogeneous mixing and fabricated into the wafer. We investigated the release pattern of PD from PD/PLGA wafers and utilized these wafers in experimental aural cholesteatoma which was induced by ear canal ligation (ECL) method. We measured the number and the surface of osteoclast from the capitatum of mongolian gerbil in which PD/PLGA wafers were implanted to evaluate effect of PD. The osteoclast cell number and surface decreased in PD/PLGA treated group indicating that the sustained release of PD could suppress the osteoclstic bone resorption at the area adjacent to the experimental cholesteatoma.
