제목 Antitumor activity of BCNU loaded PLGA Wafer against 9L gliosarcoma cells in vitro and in vivo
초록 Administration of anticancer drug into the central nervous system using implantable polymeric device has been developed for treatment of malignant brain tumor. Locally controlled release of anticancer drug can provide higher and longer exposure of drug at tumor site and reduce toxicity of systemic administration. In this study, BCNU loaded PLGA wafers were prepared with or whithout exipient such as PVP or NaCl and their antitumor activities were assessed against 9L gliosarcoma cells. Inhibition of cell growth by BCNU released from wafer was evaluated by sulforhodamine B assay of cell viability. In vivo antitumor activities of BCNU/PLGA wafers were evaluated in subcutaneous tumor model on flank of mice. Tumor size reduction and median survial rate of BCNU/PLGA wafer implanted group were evaluated and then compared with those of BCNU injected group.
