Taxanes such as paclitaxel and docetaxel are clinically available for the treatment of various cancers. However, their therapeutic effects has been severely limited by poor solubility and indiscriminate toxicity in normal cells as well as cancer cells. To overcome these problems, the dextran derivatives have emerged as promising drug carriers, owing to their excellent water solubility and biocompatibility. In particular, carboxymethyl dextran (CMD) has been often used to develop polymer-drug conjugates because it has multiple functional groups for chemical conjugations. In this study, we prepared CMD-based polymeric nanoconjugates using facile chemistry, which could improve the solubility of insoluble hydrophobic anticancer drugs and selectively release the drugs in the mild acidic condition. The physicochemical characteristics of CMD conjugates were investigated using 1H-NMR, DLS, high-performance liquid chromatography (HPLC) and confocal microscopy. |
