Aminodeoxychitosan (ADC) were successfully prepared under various conditions. ADC with various degree of substitution (DS) at C-3 and C-6 were characterized by elemental analysis, FT-IR, and proton and carbon-13 NMR. ADC samples with a DS 0.91 and 1.0 did not displayed any cytotoxicity on human dermal fibroblast proliferation. In order to enhance proteolytic stability of free EGF, EGF was conjugated with ADC* (DS 1.0) by using water-soluble carbodiimide (EGF-ADC*) in the similar manner of Park, et al (2005). The effect of EGF-ADC* treated with trypsine or not on the proliferation of fibroblast was compared with free EGF and EGF-chitosan conjugates (EGF-CS) in vitro. The cell proliferative effects of the conjugates treated with trypsine increased in the following order: Control (free EGF) < EGF-CS < EGF-ADC* and the cell proliferative effects of the conjugates treated with no trypsine were almost similar to Control’s. This result suggests that EGF may be more stabilized when conjugated with ADC* than CS. EGF-ADC* may be used as a wound healing agent. |
