Anti-wrinkle effect of EGF(epidermal growth factor) has been reported by previous research. Also, it has been demonstrated by previous our study that half-life of EGF can be prolonged by conjugating with LMC(low molecular weight chitosan). Based on these reports, we prepared EGF-LMC conjugates by using carbodiimide, and its effects on the proliferation and ECM synthesis of NHDF(normal human dermal fibroblast) were estimated as the in vitro test for anti-winkle agent. The proliferation rate of NHDF was measured by MTT assay. The result showed that the stimulatory effect of EGF-LMC on the fibroblast proliferation was stronger than pure EGF. Also, amounts of ECM synthesized by NHDF were quantified by ELISA method. The result showed that EGF-LMC treatment caused the increase of ECM synthesis compared to pure EGF treatment, particularly when higher dose were used. By these results, it is suggested that EGF-LMC conjugates can be used as anti-wrinkle agents. |
