제목 Enhanced gene transfection efficiency of chitosan for gene therapy
초록 Chitosan is a well-known drug or gene delivery vector and antimicrobial agent. Amino acid-grafted chitosan (AAC) was chemically conjugated by coupling reaction. Chemical characterization of AAC was performed by 1H nuclear magnetic resonance (NMR) and fourier transform infrared spectroscopy (FT-IR). The particle size and zeta potential of AAC were characterized by using DLS. The result of gel retardation assay showed that the AAC was effectively complexed with pDNA at low weight ratio. In addition, transfection efficiency of AAC was higher than free-chitosan in HEK 293. These results suggest that AAC is useful as a gene delivery carrier.
