제목 Production of 3-hydroxypropionic via malonyl-CoA pathway from acetate
초록 Acetate had been previously utilized as a feedstock for the production of biochemical or biofuel since it could be transformed into acetyl-CoA. In this study, production of 3-HP from acetate via malonyl-CoA pathway was attempted by overexpressing acetyl-CoA carboxylase (acc) and dissected malonyl-CoA reductase mcr (mcrC and mcrN). The overexpression of acc, mcrC and mcrN in E. coli BL21(DE3) showed 3.2 mM of 3-HP production under shake flask cultivation. The down regulation of fatty acid biosynthesis by the addition of cerulenin to culture media exhibited 2 fold enhanced 3-HP levels. However, the inefficient acetate consumption, relatively lower cell growth and reconversion of acetyl-CoA to acetate could be the major hurdles. The strategies to enhance acetate consumption rate, to enhance the growth, to minimize the acetyl-CoA consumption by competing genes and to replace expensive cerulenin are under progress to enhance the production of 3-HP from acetate.
