Multi-functional biomaterials based gene therapy could be utilized to enhance therapeutic effects (i.e. targeted delivery). Recently, adeno-associated viral vector (AAV) conducted biomaterials have been accelerating of researches, due to the final approval of commercialization by the food and drug administration (FDA, USA) and European medicines agency (EMA, EU). Herein, we developed the surface modification technique for interactions with any AAV serotypes. The recombinant AAV capture protein were engineered insertion of cysteine for maleimide-thiol “Click” chemical conjugation and produced for AAV conducting biomaterials. The tuning interfaces for AAV were showed that the significantly enhanced AAV capture efficiency and in vitro/in vivo controlled AAV transgene expression with any biomaterial structures (e.g., microspheres, nanofiber). This AAV specific functional biomaterial showed high potential for using advanced gene therapies (e.g. localized gene therapy). |
