제목 | The Injection of Muscle Derived Stem Cell/Alginate Mixture as a Treatment in Urinary Incontinence |
초록 | Many bulking agents were used for the treatment of urinary incontinence but their volume and size decreased according to the time. Also, although cell therapy could improve the function, it could not make a big bulk, where we want to make a bulk, without the biopolymer holding effect. In this study, we isolated muscle derived stem cell from rat using modified preplate method, and we evaluated that the muscle derived stem cell/alginate mixture couldbe used as a bulking agent and alginate could provide a good environment for muscle cell survival. We isolated myogenic cells from the Gastrocnemius muscles of Sprague-Dawley rats by modified preplate method and then, purified the muscle stem cells through FACS sorting and immunostaining (CD34, Sca-1, CD45 & desmin). We injected alginate with and without muscle derived stem cells into the rat bladder. After 4 weeks, the rats were sacrificed and their bladder was harvested (H&E staining, immunostaining & confocal microscopy) We modified preplate methods and isolated CD34+, Sca-1+, CD45- and desmin(-) muscle derived stem cell in rat. We think this muscle derived stem cell may have a more primitive characteristics than satellite cells. We easily injected the muscle derived stem cell/alginate mixture into the rat bladder and they changed from sol to gel state. Without cells, alginate gel was decreased the volume in spite of smooth muscle cell ingrowth into the alginate gel, but with cells, there was no volume change up to 4 weeks. We thought that either the alginate would provide a good environment for muscle cell survival, or muscle derived stem cells were immunologically inert and can be differentiated to smooth muscle cells.
