제목 Immunosensing System for Biomarkers based on Reflectometric Interference Spectroscopy
초록 Reflectometric interference spectroscopy (RIfS) is a useful approach to creating an optical biosensor, since an inexpensive silicon nitride-coated silicon substrate (SiN chip) can be used, on which the surface modification is easy to perform by using silane coupling reagents to biomolecules. The RIfS system provides real-time information on biomolecular interactions without labeling, where optical thickness was monitored on the sensor surface due to the change of physical thickness and/or refractive index caused by the binding of analytes. In this study, we developed a RIfS-based immunosensing system for C-reactive protein (CRP). In order to increase the sensitivity for the target proteins, the oriented immobilization of antibody was performed. A SiN chip surface was modified by carboxymethyl dextran and then, CRP antibody was immobilized via protein A on the CMD modified SiN chip. The typical sensitivity of the RIfS-based immunosensor toward CRP was found to be 100 ng mL-1.
