Nonvascualr drug eluting stents(DESs) was medical devices for treatment of gastrointestinal cancers such as esophageal, bile duct, ureteral, prostatic. Hydrophilic drug, gemcitabine(GEM) was investigated and reported for their potential for local treatment of cancers. But, the effect of cancer was limited because of Gem has a high hydrophilic property. So, Gem was not penetrated effectively to cancers. To solve this problem, penetration enhacer was incorpotated to hydrophilic drug eluting stent. The polyurethane, Gem, penetration enhacer was dissoved in organic solvent and then, coated in polytetrafluoroethylene(PTFE)-covered stent. The therapeutic effect confirm in cell study using multilayred cell sheet model. Also, confirm therapeutic effect in mouse model which, induced bile duct cacner in back skin. Consequently, penetration enhancer was enhancing of hydrophilic drug to cancers, and thus, it has potential for local cancer therapy. |
