Thermogel for stem cell spheroid formation in situ |

Thermogelling polymers of PEG-PPG-PEG and pyridine-dicarboxylate underwent sol-to-gel transition as the temperature increased. The gel modulus at 37 oC could be controlled in a range of 800-8,000 Pa by adding Fe3+ to the polymer aqueous solution. Transmission electron microscopy images exhibited changes in nanoassemblies from fibers to spheres, and to ribbons as the Fe3+ concentration increased. Tonsil-dereved mesenchymal stem cells were incorporated in the thermogel by increasing the temperature of the cell-suspended polymer aqueous solutions to 37 oC. The embeded stem cells in the in situ formed gel aggregated to form living cell spheroids at day 7. The high molecular weight of PEG and appropriate gel modulus, and cyto-friendly microenvironments might contribute to the formation of live cell spheroid in situ. Diffrentiation of the stem cells as cell spheroids into chondrocytes, osteocytes, and adipocytes were investigated. |
