제목 Formation of self-assembled monolayer of avidin crystals by molecular recognition: A new synthesis route of biotin derivative
초록 Sulfur-based molecules containing biotin have been used to create a wide variety of self-assembled monolayer (SAMs) on gold surfaces. Among various biotinylated thiols, 12-mercaptododecanoic-(8-biotinoylamido-3,6-dioxaoctyl) amide has been widely used because it forms closely packed, well-organized SAM monolayer on gold. The yield of biotin derivative is very poor (3%) when it is synthesized by the method originally proposed. We report here a new synthesis route of biotin derivative by using a protecting technique and a selectively ion-exchange chromatography method (RESIN-CAPTURE METHOD) which makes the workup steps very simple and convenient. This route is expected to increase the final product yield more than that of the original method. The final goal of the present experiments is to link the avidin microcrystals with the biotin monolayer self-assembled on metal substrate. Our study may provide an insight on mass production of protein-chip by multiplying the target protein similar to DNA polymerization in commercialized DNA-chip.
