제목 | Microbial production of actinocin, an actinomycin D chromophore, using Escherichia coli |
초록 | Non-ribosomal peptides (NRPs) are a class of natural products that possess numerous pharmaceutical applications, and contribute to billions dollars of market in medical sector. One such NRP is FDA-approved anticancer actinomycin D. In this study, an effort was made to produce actinocin, a chromophore of actinomycin D, by using E. coli as a heterologous host. L-tryptophan degradation pathway in E. coli was removed to direct L-tryptophan towards the aerobic pathway of tryptophan metabolism. Through the incorporated aerobic pathway, tryptophan was metabolized to produce actinocin in E. coli. Additional metabolic engineering strategies were further employed to enhance the intracellular L-tryptophan pool and cofactor availability of the host strain. Optimization of the tryptophan pathway together with the addition of actinocin biosynthetic pathway led to the de novo production of actinocin from glucose. The resulting E. coli strain will serve as a platform strain for the enhanced production of actinomycin D and other phenoxazine ring-containing antibiotics.
