Functional expression of ChCODH by co-expression of Suf system in engineered Escherichia coli under microaerobic conditions |

CO dehydrogenase (CODH) is a key enzyme that catalyzes CO to CO2. To utilize CODH in a two stage biocatalytic CO conversion system (CO-CO2-formate), ChCODH which originates from Carboxydothermus hydrogenoformans was overexpressed in E. coli. However, this enzyme has a disadvantage in that a functional expression was fulfilled when the iron-sulfur cluster is bound to ChCODH under strict anaerobic conditions. Due to the difficulty of maintain anaerobic conditions in biological process, we tried to performed the functional expression of ChCODH under aerobic or microaerobic conditions by introducing the Suf system that provide iron-sulfur clusters under oxidative stress conditions. As a result, the activity of ChCODH was observed under aerobic conditions, but specific activity was lower than that when ChCODH was expressed under anaerobic conditions. However, the specific activity of ChCODH was increased up to 1000 U/mg ChCODH by co-expression of Suf system under microaerobic conditions. Therefore, this study suggested the possibility of functional expression of ChCODH under microaerobic and aerobic conditions. |
