This work introduces poly-ion complexes between protein (epidermal growth factor, EGF) and liposome (1,2-dioleoyl-3-trimethylammonium-propane, DOTAP) with multi-lamellar structures via electrostatic attraction. The membrane fusion of the cationic DOTAP liposomes in neutral solution was mediated by the anionic EGF (isoelectric point = 4.6) in a layer-by-layer manner, resulting in the intercalation of the EGF into the interstitial space of multi-lamellar structures (proteolipoplexes). The average diameter (± s.d.) was about 171.6 ± 4.3 nm, and the loading efficiency of EGF was higher than 90 %. In vivo percutaneous penetration of the EGF from the proteolipoplexes was greatly higher than pristine of EGF indicating lipid-mediated integration. Acknowledgement: Celltrion, Inc.
