Human serum IgG shows long circulating half-life (~ 3 weeks) in sharp contrast to other protein therapeutics and its lifespan is highly variable depending on the interaction of Fc domain and FcRn. Association of IgG Fc domain into FcRn at slight acidic endosome and dissociation of the Fc-FcRn complex in serum allow intracellular trafficking of IgG antibodies and recycling, which in turn leads to prolonged serum half-life of IgG antibodies. To further improve the circulating half-life of therapeutic IgG antibodies, a variety of biomolecular, cellular and evolutionary approaches have been employed and will be discussed. |
