제목 | Development of RNA based sensor with riboswitch for monitoring high-value added product, naringenin |
초록 | Detection and measurement of intracellular high-value-added products can improve productivity from microbial cells. RNA based genetic regulators, the riboswitches are can be used to modulate the expression of genes in response to their cognate ligands. The characteristic of riboswitch makes it possible to facilitate the detection or selection of target products. Here, we developed artificial riboswitch, which can control gene expression by high-value-added product, naringenin through an abridged process. This process involves the modulation of naringenin concentration and selection pressure level during negative and positive selection. After the entire process, we develop naringenin riboswitch that has been improved in specificity against other flavonoids. We expect that these new riboswitches could be applied to diverse applications in metabolic engineering and investigation of flavonoid metabolism. |
