The goal of this study is to research effect of gellan gum for bio disc tissue engineering on the cell viability and extracellular matix secretion and to select the best gellan gum of contents for nucleus pulposus to grow. The preparation of gellan gum gels was designed by varying the gellan gum contents with 1, 2 and 3 wt% and cross-linked by EDC. NP of rabbits was cultured in vitro for 3 weeks. Cell viability (MTT), glycosamino-glycan (GAG) assay, reverse transcription polymerase chain-reaction (RT-PCR), hematoxylin-eosin (H&E) and safranin-O stainining were evaluated to analyze the effect of the gellan gum on the cell viability and extra cellular matrix secretion. These results demonstrated that NP prefer the gellan gum with the higher the concentration of gellan gum for better proliferation and ECM production. It is believed that the gellan gum would significantly affect the cell metabolism. This research was supported by WCU (R31-20029) and MBC (0405-BO010204-0006) . |
