Purification of protein using Ni2+- Poly(2-acetamido acrylic acid)PAAA hydrogel developed for Immobilized Metal Affinity Chromatography(IMAC) |
| 초록 |
We developed novel protein purification system using metal-complexed poly(2-acetamido acrylic acid)(PAAA) hydrogel based on immobilized metal affinity chromatography(IMAC). Recombinant Histidine tagged green fluorescence protein(His6-GFP) was purified using the developed method. Confocal laser scanning microscopy(CLSM), Photoluminescence(PL), and ssodium dodecyl sulfate-polyacrylamide gel electrophoresis(SDS-PAGE) analysis clearly showed that His6-GFP was selectively bound to Ni2+-PAAA hydrogel. This developed method has several advantages including low cost and a simple purification procedure. |
