제목 Self-assembled nanostructure based on  M13 bacteriophage Color Sensor for Stem Cell Differentiation Monitoring
초록 In stem cell in vitro amplification culture, in order to determine the cell behavior such as proliferation, aging, differentiation of cells, expensive equipment, experimental techniques, and a lot of time is required to confirm this. However, in the case of diseases such as heart disease and cancer, the prognosis of the disease worsens over time, so it is essential to monitor the functional cell state applied to the treatment in real time. The cell we used for monitoring is CPC(Cardiac progenitor cell). CPC is capable of self renewal and differentiation into three cardiac lineages: cardiomyocyte, smooth muscle cell, and endothelial cell. In this regard, cell transplantation therapy using CPC has been suggested as a treatment strategy for cardiovascular diseases for impaired myocardial regeneration.
Colorimetric sensors can be easily fabricated by self-assembly of M13 bacteriophage by genetically engineering peptides on surfaces. Twenty amino acids have different characteristics (Hydrophobic, Hydrophilic, Aliphatic, Sulfur, Hydroxyl, Acidic, Basic, Hydroxyl, polar, Charge) and can be genetically manipulated to produce different multi-array-based selective electronic nose sensors. This sensor system was used to analyze the color change pattern caused by the reaction between the sensor array and the foreign material. Differentiated cell respiration was applied to the sensor array to analyze the color change pattern, and the results were separated by specific hierarchical cluster analysis. It is anticipated that our biosensor systems could be used to develop assay kits that are inexpensive, easy to supply and applicable to cell monitoring.
