Freeze-dried cell-free in vitro transcription and translation system enables rapid and inexpensive expression of recombinant proteins at any site. Using genetic devices that can sense nucleic acids, on-site molecular diagnostics on the portable protein expression platform can be created. In this talk, I will introduce the freeze-dried in vitro transcription and translation platform adopted to develop programmable molecular diagnostics. In combination with genetic devices, a femtomolar or lower level of viral sample is identified in a simple, and field-ready workflow based on in vitro transcription and translation as a protein expression basis. Overall, the given cell-free biological platform that resolves important practical limitations facing many different applications demonstrate how cell-free synthetic biology can be used to develop future molecular devices for molecular diagnostic tools. |
