Preparation of basic proteinoid included cubic phase containing hydrophobically modified glucose oxidase for glucose-sensitive release |

Basic proteinoid included monoolein (MO) cubic phase containing hydrophobically modified glucose oxidase (HmGOD) was prepared for a pH-dependent and glucose-sensitive drug carrier.The basic proteinoid was prepared by condensation of Arginine and Leucine. When pH > pI, the basic proteinoid was condensed as shown in the absorbance determined by UV spectruphotometer. Then mixture buffer solution of basic proteinoid and HmGOD was used to hydrate the molten MO to prepare cubic phase. The release of crystal violet from the cubic phase was promoted under high concentration of glucose conditions. This might due to HmGOD interaction with glucose that reduce pH in water channel of cubic phase which would further affect the morphology of basic proteinoid and trigger high release. Moreover, with changing pH of the buffer solution in water channel would trigger high release. Therefore the releases of crystal violet from MO cubic phase could responsive to both glucose concentration and pH. |
