제목 L-ornithine production in Corynebacterium glutamicum through metabolic engineering
초록 L-ornithine, a non-essential amino acid has been used in food industry. Here, we engineered Corynebacterium glutamicum ATCC 13032 to produce L-ornithine by metabolic engineering methods. To modify metabolic pathway, we manipulated bacterial genes and operon system. Especially, to enrich the NADPH pool, we changed the start codons of the pgi and zwf genes and altered the original promoter of the tkt operon. As a result, the final strain showed production of 51.5 g/L of L-ornithine in 40 h with productivity of 1.29 g/L/h in fed-batch cultivation. This results represent how manipulated C. glutamicum can produce L-ornithine in effective way.
