In our previous study, meniscal cell/polycaprolactone (PCL) hybrid scaffolds were prepared and characterized. The PCL scaffold exhibited high porosity and showed similar mechanical properties as commercialized scaffolds for meniscus regeneration, such as CMI®. From the in vitro cell culture, it was observed that the meniscal cells were uniformly grown inside the PCL scaffold. In this study, meniscus-shaped PCL scaffolds with and without meniscal cells were implanted into New Zealand White rabbits to evaluate the meniscus regeneration from the animal. The meniscal cells were seeded into the PCL scaffold, cultured in vitro for 2 weeks, and transplanted to rabbit knee joint from which whole medial meniscus was removed. After 1, 3, and 6 months transplantation, the implant specimens were evaluated by histological (H&E, Safranin-O, and M-T staining), biochemical (water, collagen content), and mechanical (compression test) studies. |
