Effect of Silk Contents on the Attachment and Proliferation of Retinal Pigment Epithelial Cells on Silk/PLGA Films |

Biomaterials for retinal tissue engineering must demonstrate several critical features for potential utility, mechanical integrity, biocompatibility and slow biodegradation. Silk is being rediscovered and reconsidered as potentially useful biomaterials in a range of applications. We prepared natural/synthetic hybrid silk/PLGA films using 0, 10, 20, 40 and 80 wt% of silk by the solvent evaporation method. We measured MTT assay to confirm cellular of cell number attached on film at 1, 2 and 3 days. Morphology of cellular adhesion on films was confirmed by scanning electron microscope (SEM) at 1, 2 and 3 days. RT-PCR was conducted to confirm mRNA expression of RPE65 as RPEs marker. The expression of cytokeratin was determined by immunofluorescence staining. We confirmed that specific 20~40 wt% silk in silk/PLGA film was superior to adhesion and proliferation of RPEs.
