제목 Effect of Silk Contents on the Attachment and Proliferation of Retinal Pigment Epithelial Cells on Silk/PLGA Films
초록 Biomaterials for retinal tissue engineering must demonstrate several critical features for potential utility, mechanical integrity, biocompatibility and slow biodegradation. Silk is being rediscovered and reconsidered as potentially useful biomaterials in a range of applications. We prepared natural/synthetic hybrid silk/PLGA films using 0, 10, 20, 40 and 80 wt% of silk by the solvent evaporation method. We measured MTT assay to confirm cellular of cell number attached on film at 1, 2 and 3 days. Morphology of cellular adhesion on films was confirmed by scanning electron microscope (SEM) at 1, 2 and 3 days. RT-PCR was conducted to confirm mRNA expression of RPE65 as RPEs marker. The expression of cytokeratin was determined by immunofluorescence staining. We confirmed that specific 20~40 wt% silk in silk/PLGA film was superior to adhesion and proliferation of RPEs.
