제목 Mechanism based development of Dicer Substrate RNA for potent in vivo RNA interference
초록 Various RNA binding proteins are involved in RNA interference(RNAi). Our previous study showed Dicer substrate RNAs enhanced RNAi potency over conventional siRNAs. Using FP analysis, we have identified the interaction of Dicer substrate RNA with proteins to further understand RNAi mechanism. The biological properties of Dicer substrate RNA need to be improved for clinical applications. Chemical modification is effective strategy to reduce nuclease degradation and immunogenicity. Alternating or sequence-substitution are conventional modification approaches. However, modification site should be carefully selected because modification on specific position could disturb critical interaction with RISC proteins. Therefore, we developed protein-RNA interaction(PRI) based modification. New position specific modification(PSM) can be applied to various Dicer substrate RNAs with the wide range of target sequences. Finally, Dicer substrate RNA with PSM showed more potent in vivo gene silencing.
