제목 In Vitro and In Vivo Gene Delivery Using Branched Poly(Ethylene imine)-graft-Carboxymethyl Chitosan Copolymer
초록 We conjugated O-Carboxymethylation Chitosan (OCMCh) with Poly(Ethylene imine) (MW 25,000) (PEI). To enchance the low tranfection efficiency of chitosan and reduce the high cytotoxicity of PEI. The physicochemical characterisitc of OCMPEI was investigated by 1H-NMR. Particle size distribution of the OCMPEI/pDNA complexes nanoparticles, were determined 410 ~ 195 nm using by the dynamic light scattering(DLS) technique. Gel electrophoresis of the OCMPEI/pDNA complexes were confirmed that binding affinity, pDNA protection affinity and release of pDNA. As a results, OCMPEI was good biding affinity, good protection capacity of pDNA. OCMPEI 15% was best of transfection efficiency of pDNA in cancer cell line by fluorescence microscopy.
