we developed a novel method that decreases particle size of drug through a simple process using ‘Porous Sugar Template (PST)’. PST was prepared by directional freezing and subsequently freeze-drying a solution of mannitol and lactose in water (50 wt%). After preparation of PST, drug solution (naproxen in acetone, 1 wt%) was transferred to PST. Inside PST pores, drug was crystallized. Besides, the pores constrained the growth of the crystals. Using the Mie-Fraunhofer light scattering method, it was confirmed that the particle size of drug crystals decreased down to 500 nm from 10 ~ 20 um. However in another approach using a porous template prepared by directional freezing and freeze drying a solution of mannitol and drug in a mixture of water and acetone, the crystallization of drug was not effectively controlled. This process results in a mean diameter of 8.7 um including 42 % of nanoparticles. This study showed that the pores of PST had a significant confinement effect on the crystallization of drug. |
