The structural protein elastin is one of the main components of the extracellular matrix, which provides structural integrity to the tissues and organs of the body. Not only elastin but also its soluble forms, α-elastin, have inspired for many years because of its ability to self-assemble under physiological conditions. We present here the fabrication of insulin-loaded nano/micro-particles using PEGylated α-elastin derived from human adipose tissue. PEGylated α-elastin can be self-assembled into spherical nano/micro-particles with multicore structure exhibited a high protein loading efficiency. The self-assembled PEGylated α-elastin nano/micro-particles were able to encapsulate significant amounts of insulin by simple mixing at low temperature in distilled water and heating up to body temperature. These thermo-responsive α-elastin nano/micro-particles have potential as a smart protein/drug delivery system. |
