Chaperone assisted improvement and redirection of microbial whole cell activity |

Chaperones, the proteins assisting folding of intracellular proteins in organisms, have been generally utilized to improve the productivity of functional recombinant protein in microbial hosts such as Escherichia coli. In this presentation, it is suggested that chaperone can be employed as an assistant to improve and redirect a microbial whole cell activity. First, it is proposed that the coexpression of chaperone can improve the whole cell biocatalytic activity of recombinant strain. For example, whole cell activity of recombinant E. coli engineered to express the styrene monooxygenase (SMO) from Pseudomonas putida can be enhanced by coexpressing the GroELS chaperone with the SMO gene. Second, it is shown that the overexpression of wildtype or engineered chaperone in a target microbial host can redirect the microbial activity against temperature. |
