제목 | Chitosan Microsphere for Nasal Vaccine Delivery System |
초록 | Chitosan is a valuable excipient for nasal drug delivery systems due to its biocompatibility, biodegradability, low cost and the ability to open intercellular tight junctions. Also, it was reported that chitosan could induce cytotoxic T lymphocytes and macrophage activation. In this study, chitosan microparticles (CM) for vaccine formulation were prepared based on ionic gelation process of tripolyphosphate, and then chitosan microspheres loaded with Bordetella Bronchiseptica Dermonecrotoxin (BBDNT), a virulence factor of a causative agent of atrophic rhinitis, were prepared for nasal vaccination . The particle sizes of CM-BBDNT were smaller than 5 μm and had narrow size distribution with uniformity. The loading efficiency of BBDNT with CM was even higher (over 70%) than normal vaccine formulation irrespective of the molecular weights of chitosan (300, 100, and 10 kDa). The higher pH accelerated the BBDNT release from CM-BBDNT, indication of the strong interaction between CM and BBDNT at the lower pH environment. Also, much and longer TNFα were secreted from CM-BBDNT than BBDNT itself although both BBDNT itself and CM-BBDNT had stimulated macrophage in early stage, suggesting that the latter maintained strong immune response more effectively than the former. |
