제목 | Galactosylated Poly(4-vinyl imidazole) as Hepatocyte-Targeting Gene Carrier |
초록 | The use of non-viral gene carrier such as cationic polymers or cationic lipids may overcome the current problems associated with viral gene carrier in safety, immunogenicity and mutagenesis. In this study, poly(4-vinylimidazole) (P4V) of which the side chain is similar to that of natural amino acid, histidine, was used as a gene carrier and conjugated to lactobionic acid bearing galactose group for liver specificity. The Gal-P4V/DNA complex was confirmed using agarose gel electrophoresis. The sizes and zeta potential of the complex were measured by light scattering. The cytotoxicity of Gal-P4V was evaluated. The Gal-P4V/DNA complex was transfected into HepG2 cells having asialoglycoprotein receptors (ASGR), indication of specific interaction of ASGR on HepG2 cells and galactose ligands on Gal-P4V. |
