제목 Galactosylated Poly(ethylenimine) (Gal-PEI)-graft-poly(vinyl pyrrolidone) (PVP) as Hepatocyte-Targeting Gene Carrier
초록 The use of non-viral gene carrier such as cationic polymers or cationic lipids may overcome the current problems associated with viral gene carrier in safety, immunogenicity and mutagenesis. In this study, polyethylenimine (PEI) was used as a gene carrier and conjugated with lactose bearing galactose group for hepatocyte targeting and grafted with poly(vinyi pyrrolidone) (PVP) as a hydrophilic group to reduce cytotoxicity of PEI. The Galactosylated (Gal) PEI-PVP (Gal-PEI-PVP)/DNA complex was confirmed using agarose gel electrophoresis. The sizes and zeta potential of the complex were measured by dynamic light scattering. The cytotoxicity of Gal-PEI-PVP in HepG2 cells was evaluated. The Gal-PEI-PVP/DNA complex was transfected into HepG2 cells having asialoglycoprotein receptors (ASGRs), indication of specific interaction between ASGRs on HepG 2 cells and galactose ligands on Gal-PEI-PVP
